Tylosin extracted from fermentation broth is a macrolide antibiotic. At present, the conventional extraction process is generally to pre-treat the tylosin fermentation liquid and filter it to obtain the fermentation filtrate, then extract the fermentation filtrate with ethyl acetate to obtain the tylosin-rich organic phase, and then back-extract it with acid water, and then obtain the tylosin tartrate or tylosin phosphate fine product after neutralization, decolorization, concentration and drying.
In industrial production, enterprises generally need to improve the above common extraction and purification process. Chloroform or dichloromethane can be used to extract tylosin from the fermentation filtrate. The impurity extraction rate is very high, and a large amount of red pigment and other impurities can be extracted. However, it is difficult to remove some impurities in the later stage, which will seriously affect the quality and yield of tylosin.
Zhengzhou Tiei Extraction, in response to this problem, proposed a technology combining the composite extractant with CWL-M three-stage countercurrent centrifugal extraction to optimize the tylosin purification process and effectively improve the quality and yield of tylosin products. Among them, the composite extractant is composed of ethyl acetate-chloroform (or trichloroethane)-petroleum ether. After three-stage countercurrent extraction by CWL-M centrifugal extractor, the purity and quality of tylosin products are greatly improved, and the total tylosin components of the extract are not less than 98%.
